        abstract:  This study estimated the concentration of microplastics (i.e. plastics measuring less than 5mm; reported in unit of count per individual and count per gram) in Scylla Serrata (Giant mud crab) from the Kota mangroves, southwestern coast of India collected during 2022-11-25. Microplastics were collected using a cylindrical crab trap. Percentages of microplastics in various size ranges and types are reported. This dataset contains the results from all 9 giant mud crab samples, in a spreadsheet format.

                description:  Parameter or Variable: microplastic concentration (measured); Units: count per individual; Observation Category: in situ; Sampling Instrument: cylindrical crab trap; Sampling and Analyzing Method: Nine samples of the mud crab species (Scylla serrata) were collected from a fisherman near the Kota mangroves. The fisherman had collected the crab samples with the aid of a cylindrical crab trap which were deployed within the mangroves. The samples were neatly packed in aluminum foil and kept in steel containers to avoid contamination. They were safely transported to the laboratory and stored in a deep freezer until further analysis. Prior to sampling, ethical clearance was obtained from the Institutional Animal Ethics Committee of the Manipal Academy of Higher Education to undertake this study. Before dissecting the mud crabs, the length and width of their carapaces, weight of the whole body were measured. Based on their length and width measurements, all the samples belonged to the adult category. Later, these samples were thoroughly rinsed using pre-sieved (0.1 mm) double distilled water to remove any external contaminants. The carapace was removed, and the gastrointestinal tract (GIT) was carefully dissected with the help of a stainless-steel knife and scissors of surgical grade. The weight of the gastrointestinal tract was noted. The digestion of the biological matter was conducted in two phases. In the first phase, potassium hydroxide (10 %) was added to inside a laminar flow to avoid airborne contamination. After the biological matter broke down, the samples were dried at 50 ◦C and later treated with hydrogen peroxide (30 %) for complete digestion of the biological matter. The clear solution was passed through four sets of sieves (5 mm, 1 mm, 0.3 mm, and 0.1 mm stacked one above the other with the largest mesh size on top and the smallest at the bottom). The residue or material present on the 1 mm, 0.3 mm, and 0.1 mm sieves was carefully transferred to watch glasses and considered for the identification of MPs. Microplastics were identified using a stereo zoom microscope (NIKON SMZ-745-T) at 40× magnification. Under the microscope, parameters such as abundance, shape, size, and color were studied. Only those particles meeting the following criteria were considered MPs: particles that lacked any cellular or organic structure, had uniformly thick fibers, did not break upon prodding with tweezers, changed behavior during a hot needle test, and stained with 1 % Rose Bengal. The polymer composition of 95 microplastics (0.3–5 mm size fraction) was determined using Fourier transform infrared spectroscopy with attenuated total reflectance (FTIR-ATR; Shimadzu IRSpirit). The selection of these 95 microplastics was made to ensure that all the different types of microplastics observed during visual identification can also be characterized by IR spectroscopy. Percent transmission was recorded across a spectrum of 3500 to 500 cm-1 using a resolution of 4 cm-1. Prior to scanning, the ATR crystal was carefully cleaned with ethanol, after which the MP was scanned around 25 times. The IR spectra obtained for the samples were matched with reference spectral curves on Open Specy, an open-source software for identifying spectral signatures of polymers (Pearson correlation coefficient > 0.8). The surface morphological features of four fibers which were frequently observed in the gut of the crab were studied using a scanning electron microscope (EVO MA18) at the Innovation Centre, Manipal Institute of Technology.; Data Quality Method: The analysis was conducted in a clean laboratory setting with minimal disturbance from other personnel. All solutions used in the analysis were prepared using double-distilled water (DDW). Only stainless steel and glass apparatus were used during the analysis. All beakers were thoroughly cleaned with DDW and covered with aluminum foil to avoid atmospheric contamination. All the liquids, such as DDW, and all solutions used during analysis were pre-sieved to avoid any external contamination. Nitrile gloves and cotton laboratory coats were used throughout the duration of the experiments. Four laboratory blanks were kept to check the atmospheric contamination inside the laboratory..
