description: Parameter or Variable: microplastic concentration (measured); Units: pieces/m3; Observation Category: in situ; Sampling Instrument: Stainless steel spoon; Sampling and Analyzing Method: Playa Grande beach (Tenerife, Canary Islands, Spain) was sampled during a complete moon cycle between 17th June and 16th July 2019 following the indications of the Guidance on Monitoring of Marine Litter in European Seas. Sampling was developed at the high tide line and with a separation of approximately 10m between each of the 10 sampling points. Briefly, a wooden frame of 50×50 cm was used to delimitate the sampled sand fraction. A stainless-steel vessel was used to collect the sand down to 5 cm deep, which was sieved in-situ using 1- and 5-mm stainless-steel circular sieves of 20 cm of diameter. The collected fraction in each sieve was introduced in glass bottles and/or Petri dishes and taken to the laboratory for further analyses. Collected microplastics were separated by density using a saturated NaCl solution (the approximate density of the solution was 1.2 kg/L). Afterwards, the floating material was passed through a paper filter using a Bücher funnel. The filtrate was then washed with deionized water and air dried. Non-plastic materials such as pieces of algae, tar, wood, etc. were then removed while plastics were weighted (Sartorius Entris 224I-1S analytical balance with a maximum weighting capacity of 220 g and 0.1 mg of resolution). The collected microplastics (1-5 mm) were counted and visually characterized by size and shape. Concerning mesoplastics (5-25 mm), they were also introduced in the NaCl solution to verify their flotation, cleaned with deionized water, dried and also weighted.; Data Quality Method: The plastics were confirmed using infrared technology. For infrared (IR) measurements a Fourier Transform IR (FTIR) spectrometer - Attenuated Total Reflection (ATR)-, Bruker IFS 66/S equipped with a high-intensity global source, KBr beamsplitter and a deuterium triglycine sulphate (DTGS) detector was used. Briefly, the instrument was operated in double sided, Forward-Backward mode, with a speed of 10 kHz. FTIR spectra were collected over the wavenumber range of 4500-500 cm−1 at a resolution of 8 cm−1 and at a rate of 16 scans per sample. OPUS 6.5 software was used to acquire and analyze data. The number of repetitions was adjusted for every sample depending on signal-to-noise ratio and the quality of spectra. A minimum of three spectra were taken per particle at random points over the particle surface. Polymer identification was performed by using the spectral libraries included in OPUS 6.5 spectra software, using the point-to-point correlation algorithm after applying a standard normalization and Savitzky-Golay first derivative to the complete spectra. The minimum matching for positive identification was set at 700 over 1000 hit quality. Additionally, the library was tested before microplastics identification, using fragments of commercial well-labelled polymers of polyethylene terephthalate (PET), high-density polyethylene (HD-PE), low-density polyethylene (LD-PE), polypropylene (PP), polyvinyl chloride (PVC), polystyrene (PS) and polyamide -nylon- (of 1-2 mm size), obtaining 100 % of positive identification, always over the 70 % previously mentioned. Spectra under 700 quality hits where visually checked, re-processed for an adequate wavenumber interval if necessary (removing possible artefacts from spectrum) and re-classified through the library. Those remaining under 700 quality hits were classified as non-identified..
